Pancreaticcancerisafataldiseasewithlowsurvivalandhighmortalityrate,whichisconsideredasignificantpublichealthchallenge.DuetotheresistanceofpancreaticcancertorADIotherapyandchemotherapy,newtreatmentapproachesareurgentlyneeded.PreclinicalstudiesofthePGHAM-1celllinecanhelptounderstandtheetiologyofthediseasebetteranddiscovernewtherapiesforpatientswiththislife-threateningcondition.PGHAM-1celllinewasderivedfromanN-nitrosobis(2-oxopropyl)amine(BOP)-inducedpancreatictumorofaSyrianhamster(Mesocricetusauratus).Thetumorsweremincedandsubcutaneouslytransplantedwithatrocarintotheinterscapularareaofuntreated5-week-oldhamsters.Therecipienthamsterswerekilledat6–8weeksaftertransplantation,andaportionofthetumortissuewasseriallytransplanted.Aftersubcutaneoustransplantation,thetumorwasextractedundergerm-freeconditions.Cellsthathadbeenmaintainedinculturefor60passagesandhadbecomeestablishedasacelllinewerenamedPGHAM-1.Thetumorshowedastrikingsimilaritytohumanpancreaticductaladenocarcinomaandhadamoderatetowell-differentiatedhistology.PGHAM-1containsaK-raspointmutationatcodon12,oneofthemostfrequenttypesofK-rasmutationinhumanpancreaticcancers.ResearchershavefrequentlyusedPGHAM-1inexperimentsbecauseoftheircells’rapidgrowthandahighrateofmetastasis.PGHAMcellsarebeneficialinbiomedicalstudiesrelatedtopancreaticcancer.ThehamsterpancreaticcancercellsofthePGHAMcelllinegreatlyresemblehumanpancreaticcancercellsandarehenceusefulincancerresearch.AltogenBiosystemsprovideshigh-efficiencytransfectionkitsforthePGHAMcellline.
